丁咯地尔对大鼠坐骨神经嵌压伤后背根神经元凋亡及血管内皮生长因子表达的影响
Effects of buflomedil on neural cell apoptosis and expression of vascular endothelial growth factor in dorsal root ganglionic following crushing lesion of sciatic nerve in rats

Abstract
Aim: To explore effect of buflomedil on the apoptosis of the neuron and the expression of vascular endothe-lium growth factor( VEGF) in dorsal root ganglionic( DRC) following crushing lesion of sciatic nerve in rats. Methods:A total of 84 SD rats were divided into the normal group, model group [normal saline,0.8 mL/( kg ? D)],and buflomedil group [40mg/(kg ? D) ] in random,Drugs were given to each group except normal group by intrapritoneal injection once per day for 14 days, then the sciatic nerve lesion models were performed, the apoptosis of the neuron cells was detected u-sing the TUNEL methods,and the expression of VEGF was detected by immunohistoehemical staining at the end of 6 h, 12h, 24 h, 48 h, 72 h, 96 h, 7 days,respectively. Results:There was less TUNEL positive cells and expression of VEGF in normal group; at the end of 12 h after the model perfomed, the apoptosis of the neural cells in model group increased obviously than the buflomedil group, peaked at 72 h after the sciatic nerve lesion, and it decreased at the end of 7 days( F = 13. 790,P<0. 001 ) ; and also the apoptosis of the neural cells in buflomedil group increased, compared with the model group at the end of 12 h, decreased obviously at the end of 24 h - 7d, compared with model group(P<0.05). There was less expression of VEGF in normal group; the expressions of VEGF increased in the model group at the end of 48 h after the model perfomed,lasted to 72 h(P<0.05) ;and also the expression of VEGF in buflomedil group increased obviously at the end of 24 h(P<0.05) .peaked at 48 -72 h, lasted to 96 h(F = 70.430,P<0. 001) , Compared with model group,the expression of VEGF were obviously different at the above times( P<0.05). Conclusion: Buflomedil could up-regulate the expression of VEGF, and performs neuro-protective effect by inhibiting apoptosis.关键词
