摘要

Abstract

BACKGROUND: Genetic modification technology of human umbilical cord-derived mesenchymalstem cells (hUCMSCs) isnumerous and varies in the complexity of their efficiency, reliability and safety.OBJECTIVE: To compare transfected efficiency of different novel chemicaltransfection .eager* on hUCMSCs.METHOOS: The hUCMSCs mere transfected with Fugene HD. Lipofectamine LTX. And Attractene. Positive cells mere detectedunder fluorescent microscope, and expression efficiencies were measured by using flow cytometry. Cell viability was measuredby using trypan blue exclusion after transfection.RESULT SAND CONCLUSION: The highest transfected efficiency was observed with Lipofectamine LT×(32.S± 2.12)%<0 05). Followed by Fugene HD (4.3±0.64)%. Minimal trarsfection efficiency mas observed with Attractene (174±0.08)%.Lipofectamine LTX demonstrated the lomer cell viability (69.8iB.3y* compared mith Fugene HD (92.4±4.2)% adAttrattene(1O8.6±3.9)% (P > OSS), but the difference mas not significant. Insummary.this study demonstrates that Lipofectamine LT×is arelative ideal chemical transfection reagent for hUCMSCs.
