Culture of human bronchial epithelial cells in vitro

Abstract
BACKGROUND: Culture of human bronchial epithelial cells has been increasingly applied in the research of respiratorydiseases.OBJECTIVE: To explore a feasible in witro culture method for human bronchial epithelial cell line supplied by American TypeCulture Collection.METHODS: Different culture conditions of human bronchial epithelial cells were tested with triate and errors. Afeajible methodwith DMEM/F12 containing 20% fetal bovine serum was finalized.RESULTS AND CONCLUSION: The cultured human bronchial epithelial cells mere flat, polygonal, and dis played acobblestone-Ike distribution when overgrown. Immunohistochemical staining results showed an expression of epithelial markerkeratin in cultured cells. The expression of nuclear factor?9. Tumor necrosis factor a and interleuVin 8 mas increased significantlyin cultured cells exposed to sputum supernatant from patients with bronchiectssis. These findings demonstrated that the culturedhuman bronchial epithelial cells mere obtained successfully. Therefore, there is no need to stick to the culture conditionrecommended by American Type Culture Collection. The modified culture method is simple, feasible and valuable.关键词
