紫外线通过诱导TNF-α胞内聚集间接调控VEGF分泌表达
Intracellular accumulation of TNF-α is involved in VEGF induction upon ultraviolet B irradiation

Abstract
Objective To explore the role of TNF-ct in mediating VEGF expression under UVB exposure. Methods The expression plasmid containing TNF-α short hairpin RNA( shRNA ) was introduced into the mouse embryonic fibroblasts ( MEFs ) and then the stable transfectants were established. The efficiency of TNF-α shRNA in blocking TNF-α induction under UVB irradiation was confirmed by RT-PCR assay. The transcription of vegf under UVB irradiation was determined by the luciferase reporter assay. The secretion of VEGF induced by UVB was determined by ELISA. Results TNF-α shRNA efficiently blocked UVB-induced TNF-α transcription. Under the same condition, VEGF transcription and expression were significantly inhibited. Conclusion TNF-ct induction is involved in VEGF expression in the cellular UVB response.关键词
