Effects of Morin on Expression of Poly Adenosine Diphosphate Ribose Polymerase and Nuclear Factor-κB in Rats Undergone Cerebral Ischemia-Reperfusion Injury

Abstract
Objective To investigate the effects of morin on the expression of poly adenosine diphosphate ribose polymerase (PARP) and nuclear factor-κB (NF-κB), both of which are inflammatory transmitters,in rats undergone cerebral ischemia-reperfusion injury. Methods The model of ischemia reperfusion injury of middle cerebral artery was induced by suture embolism method. Rats were randomly divided into sham operation group, model group, nimodipine group, and low-, middle- and high-dose morin groups. The neurological function was investigated by neurological severity scores (NSS) , and the expressions of PARP and NF-κB protein and their mRNA in infarction region were detected by using western blot and quantitative real-time fluorescence quantitative polymerase chain reaction (RT-PCR), respectively. Results Compared with those in the model group, the neurological scores (NSS) of low-, middle- and high-dose morin groups were significantly decreased (P<0. 01) ; the expressions of PARP protein, NF-κB protein, PARP mRNA,and NF-κB mRNA in penumbral area were also significantly decreased (P<0. 05,or P<0. 01).The effects of high-dose morin were significantly superior to those of low- and middle-dose morin as well as nimodipine in decreasing the levels of above-mentioned indexes (P<0. 05,or P<0. 01). Conclusion Morin can inhibit inflammatory reaction by down regulating the expressions of PARP and NF-κB; consequently, it promotes the recovery of damaged nerve function induced by cerebral ischemia injury.关键词
