Reconstruction of recombinant lentiviral vector containing human CBFα1 gene and its expression in human periodontal ligament fibroblasts

Abstract
To construct a recombinant lentiviral vector containing human CBFα 1 gene and to investigate itsexpression in human periodontal ligament fibroblasts.MethodsA lentiviral expression vector for human CBFα 1 was construct-ed by recombinant DNA technique. The reconstructed lentiviral vector containing hCBFα 1 was confirmed by PCR and sequencing. The human periodontal ligament fibroblast (PDLF) 293T cells were cotransfected with lentiviral vector pGC-FU-hCBFα 1, pHelper 1.0 and pHelper2.0 with Lipofectamine 2000. PDLFs were infected with recombinant lentiviral vector containing CBFα 1. Infection efficiency was measured by fluorescent microscope and CBFα1 expression was detected by immunocytochemical analysis.ResultsThe sequence analysis of recombinant plasmid pGC-FU-hCBFα 1 showed that the human CBFα 1 was in-serted into lentiviral vector pGC-FU accurately. The titer of concentrated virus was 2.00 x109TU/ml. Immunohistochemistry showed that CBFα 1 was expressed in PDLF 293T cells infected with recombinant lentiviral vector for CBFα 1.Therecombinant lentiviral vector pGC-FU-hCBFα 1 has been constructed and CBFα 1 are expressed in transfected PDLFs successfully.关键词
