HPLC法测定喙尾琵琶甲药材中氨基酸的含量
Determination of Amino Acids in Blaps Rynchopetera Fairmaire by HPLC
摘要
Abstract
[Objective ] The aim was to establish an HPLC method for determining amino acids in Blaps rynchopetera Fairmaire. [ Method ] After being hydrolyzed with HCl and derivating in precolumn with o-phthalaldehyde and β-mercaptoethanol, the amino acids were eluted by gradient e-lution and were detected with an UV detector (detection wavelength:338 nm). The mobile phase consisted of 0. 55% NaH2PO4( pH 7. 80) -methanol-acetonitrile,and column temperature was set at 35 ℃. [ Result]The derivatives of amino acids with o-phthalaldehyde could be separated with a stable base line within 40 min. The calibration curves for amino acids had good linearity(r =0. 997 0-1. 000 0,n =6). The average recoveries were 90.16% - 104.1% ,and RSDs were 1.89% -5.92%. [Conclusion]The method was simple,accurate and well reproduced for the determination of amino acids in Blaps rynchopetera Fairmaire.关键词
