Abstract
Objective To investigate ihe possibility of applying hyperbaric oxygen preconditioning { HBO - PC) to reduce or prevent high - altitude pulmonary edema ( HAPE) and to find out if aquaporin -5 ( AQP5) is involved in HAPE pathogenesis using a rat model. Methods Rats were divided into three groups: control, HBO and HAPE. Western blotting and real-time PCR were used to analyze (he expression of AQP5 in the lung. The wet -to - dry weighl ratio ( W/D weight ratio) and morphology of the lung were also examined. Results An increased lung W/D weight ratio was demonstrated in the HAPE group compared with the control group. Marked interstitial edema in all ling sections in the HAPE group, and small changes in 10 of the 13 sections were present in the HBO group. Lung-injury scores in the HBO group were much lower than those in the HAPE group. AQP5 in the HAPE group decreased significantly at the protein and gene level, compared with those in the control group. Protein and mRNA expression of AQP5 in the HBO group were much higher than those in the HAPE group. Lung injury in the HAPE rats was related positively to the AQP5 expression in the lung. Conclusions HBO - PC can alleviate lung injury in rats caused hy high - altitude hypobaric hypoxia and reduce HAPE incidence. HAPE in rats is associated with a downregulation of expression of AQP5 in the lung. This downregulation can be attenuated by HBO - PC. This study is the first report to introduce HBO - PC in ihe prevention of HAPE and the first investigation of the expression of AQP5 in a rat model of HAPE.关键词
