Abstract

OBJECTIVE: This study was designed to investigate the dose-dependency of XPC mRNA expression in normal human lymphoblastoid AHH-1 cells induced by 60Co γ-rays, and its possibility as a biomarker forradiation injury. METHODS: After irradiated with 0, 1, 2, 4, 6, 8 or 10 Gy of 60Co γ-rays, AHH-1 cells wereharvested at 4 h, 10 h and 24 h post-irradiation, followed by mRNA extraction, mRNA level was measured byquantitative real-time PCR. RESULTS : The mRNA expression level of XPC gene in the cells significantly increased asearly as 4 h after 0-10 Gy irradiation, and reached a first expression peak, lasting 10 h. Then, this radiation-inducedexpression continued increasing at least for 24 h and reached the second expression peak. The radiation-increasedexpression of XPC mRNA showed a dose-dependent manner within the dose range of 0-8 Gy. CONCLUSION: Theradiation-increased expression of XPC mRNA exhibited a significant dose- and time- dependency, with a potential fordeveloping into a novel radiation molecular biodosimeter.
