摘要
Abstract
[Objective] The application of One Step SYBR Green I QRT-PCR (Real-time Quantitative Reverse Transcription Polymerase Chain Reaction) in the quantitative detecting the mRNA transcription level of target gene was introduced. [ Method]The sample preparation,primer design, RNA extraction,reaction system establishment,standard curve building and quantitative gene expression during the procedure were analyzed. [ Result ] The following factors: the quality of template quality, the quality and concentration of substrate,the quality of primer, enzyme and Mg2+, and reaetion condition was key point in the success of the QRT-PCR. [ Conclusion] The reference for the pathogenesis revealing of animal disease with molecular biology technology was provided.关键词
