摘要

Abstract

[Objective] The study aimed to establish regeneration plant system of stem lump mustard somatic cell culture, to lay the foundation for genetic transformation 9ystem and biotechnology breeding of Brassicas stem lump mustard and leaf mustard. [ Method] Talcing 6 d cotyledon of stem lump mustard as materials, using orthogonal design methods, the effect of different hormone of 6-BA, NAA, 2,4-D on explants differentiation and seedling rate of callus were studied. [ Result ] The best culture medium for the growth of stem lump mustard cotyledon was MS with 6-BA 3.0 mg/L + NAA 0. 10 mg/L + 2,4-D 0 mg/L + 3 g/L agar powder; moreover, 1/2MS supplied with 0. 10 mg/L NAA was good for the rooting of tissue culture seedling. [Conclusion] The regeneration frequency of optimized medium was improved greatly.
