摘要

Abstract

To observe the effect and difference of rAd-p53 on the radiosensitivity for human hepatocellular carcionoma( HCC ). Methods Two HCC cell lines with different p53 genetic statuses, PLC/PRF/5( MT p53 ) and HepG2( WT p53 ) were infected with recombinant adenovirus carrying wild-type( WT ) p53 gene( rAd-p53 ) and Ad-EGFP, and cell survival was evaluated by MTT; Radiosensitivity was evaluated by clonogenic assay. Cell apoptosis was assayed by flow cytometry. Results 48 h after infection efficiency of 15MOI values of rAd-p53 to PLC/PRF/5 and HepG2 cells, and cell survival rates of PLC/PRF/5 and HepG2 cells were( 7. 30 ± 1. 78 )% and ( 11.51 ±1. 53 )%. The transduction efficiency was gradually increased with the infection time or virus dosage in-creased( P <0. 01 ). SER value of HepG2 and PLC/PRF/5 were 1. 24 and 1. 52 by clonogenic assay,cell apoptosis was( 22. 19 ± 1. 04 )% and( 30. 36 ±2. 74 )% respectively and the effects of rAd-p53 group was stronger than oth-ers( P <0. 01 ). Conclusion rAd-p53 can significantly inhibit proliferation, induce cell apoptosis and increase radiosensitivity of HCC, but the aforesaid efficacy of rAd-p53 in PLC/PRF/5 cells( MT p53 ) is stronger than that in HepG2 cells( WT P53 ).
