摘要

Abstract

Objective To detect the interaction of transcription factor in vivo and DNA(regions of the target gene promoters).Method The cells were fixed with Formaldehyde,the protein-DNA complex was cross-linked under the physiologic condition,DNA was broken into small fragments with sonication,the immunoprecipitation for the complex was performed with specific antibodies and heated at 65℃ to reverse the formaldehyde cross-linking,and DNA was purified by PCR.Results The fragments were amplified by using PCR to show positive bands.Conclusion ChIP is a powerful technique that endogenous transcription factors is combined DNA under different physiologic conditions.
