Quantitative Detection Method of Surface Amino Groups on Magnetic Microspheres Using Fluorescamine

Abstract

Fluorescamine quantitative detection method for detecting the density of surface amino groups of magnetic microspheres was established and the reaction conditions were optimized via the investigation of the optical reaction properties between fluorescamine and amino groups.It is found that the modified aminosilane carrying amino groups will be completely dissociated from the surfaces of microspheres after 60h under alkaline conditions,which renders a stable fluorescent signal after a further reaction of 20min with fluorescamine.It is shown that the fluorescent signal is most stable with pH of 8.0 in the reaction system.The detection method of the density of amino groups on magnetic microspheres based on fluorescamine is simple and fast with a high accuracy.
