赤眼鳟Myf5基因的克隆、序列和表达分析
Molecular Cloning and Tissue Expression of Myf5 Gene in Squaliobarbus curriculus
摘要
Abstract
The expression of the myogenic regulation factors (MRF) family determines the fate of myocytes, and controls its proliferation and differentiation. Myf5 is a member of MRF family. Total RNA of Squaliobarbus curriculus was extracted from muscle tissues. A 338bp long fragment of Myf5 cDNA in Squaliobarbus curriculus was obtained by using RT-PCR, which encodes 112 aa peptide containing partial HLH helix domain. Compared with other species, Squaliobarbus curriculus has the highest homology with Ctenopharyngodon idella. The results of tis- sue expression detected by semi-quantitative RT-PCR showed that myf5 gene was widely expressed in various tissues of Squaliobarbus curriculus, expressed much higher in kidney and heart than in gill and muscle, while it was weakly expressed in liver, brain and intestine.关键词
