摘要

Abstract

A method was established to separate DNA-protein interaction products by non-gel sieving capillary electrophoresis with laser induced fluorescence (NGS-CE-LIF). A series of effects on CE were studied, namely sieving medium concentration, separation temperature, separation voltage, pH value of Tris-Borate-EDTA (TBE) buffer. The results indicated that pUC19 DNA/MspⅠ(HpaⅡ) DNA marker and protein molecular weight marker (Broad) D532S interaction products were effectively separated when sieving medium was 0. 1% (ω/V) poly (ethylene oxide) ( 300000), electrophoresis buffer was 1×TBE (pH9. 0), separation voltage was 15 kV, and temperature was 15 ℃. The detection method of DNA-protein interaction products was used to analyze the interaction of the products of PCR of p53 gene and protein complex from gastric cancer tissue. The method has high detection rate and good separation efficiency.
