摘要

Abstract

Objective To investigate the mRNA expression and methylation of SFRP1 promoter in invasive breast cancer and adjacent normal breast tissues, and to evaluate the clinical significance. Methods The mRNA expression and methylation of SFRP1 gene in 58 paired freshed invasive breast cancer and adjacent normal breast tissues were assessed with real time fluorescence quantitative PCR and methylation - specific polymerase chain reaction ( MSP ), respectively. The correlations between these genetic features and clinical feature or prognosis were analyzed. Results The methylation of SFRP1 promoter in invasive breast cancer was significantly higher than that in adjacent normal breast tissues ( P < 0. 05 ), furthermore; so was in cancer tissues with axillary lymph node metastasis than that in non - axillary lymph node metastasis ( P <0. 05 ). However, no significant correlation was revealed between these methylation and other clinical features or prognosis. SFRP1 expression in breast carcinoma was significantly lower than that in adjacent normal breast tissues ( P <0. 05 ). In 23 cases invasive breast cancer tissues with methylation of SFRP1 promoter CpG island, the mRNA expression of SFRP1 was not detected. Conclusion Abnormal methylation of SFRP1 may down - regulate the transcription of mRNA and correlate to the oncogenesis of invasive breast cancer, thus providing effective biomarker for early diagnosis and prognosis of invasive breast cancer.
