Preservation of Macaca fasicularis sperm under low temperature

Abstract
[Objective]The present experiment was conducted to obtain the best formulation for preserving Macaca fasicularis sperm to provide basis for its cryo-preservation and quality identification. [ Method] The survival time and survival index of Macaca fasicularis sperm were determined under three temperature conditions, viz., physiological (37℃),room(15-25℃ )and low(0-5℃)temperatures. [Result]The survival time of sperm preserved in TALP solution at 37 and 15-25℃ was 12.94±5.57 and 58.59±40.91 h, respectively, and the survival index of them was 4.88±1.97 and 24.48± 17.94, respectively. The survival time and index of sperm preserved in TALP solution added with 20% egg yolk at low temperature (0-5℃) were 596.00±83.35 and 283.05±49.42 h, respectively, which were significantly higher than those of other treatments (P<0.01). [ Conclusion ]The best formulation for preservation of Macaca fasicularis sperm was recorded as TRIS solution added with 20% egg yolk at 0-5℃.关键词
