Tissue culture technology for Gynura divaricata(L.)DC

Abstract
[Objective]The present work was aimed to investigate the effects of different hormonal combinations on in vitro propagation of Gynura divaricate (L.) DC, and to develop a system for regeneration of its plantlets. [Method]The branches of G. Divaricata were used as explants to carry out induction and multiplication of adventitious buds, culture of vigorous plantlets and their rooting in culture medium containing different hormonal combinations (0-0.1 mg/L 6-BA, 0-0.2 mg/L KT, 0-0.2 mg/L 2,4-D and 0-0.4 mg/L IBA, 0-0.4 mg/L NAA). [Result]Higher concentration of 6-BA(1.0 mg/L)decreased the multiplication coefficient of cluster buds in the medium with KT and 2,4-D. Adding 2,4-D in the medium with 6-BA and KT increased the multiplication. The optimum medium for multiplication of cluster buds was MS+ 0.5 mg/L 6-BA+0.2 mg/L KT+0.2 mg/L 2,4-D which gave the highest multiplication coefficient (15.4) with strong growth of cluster plantlets. All combinations of IBA and NAA in culture medium induced rooting of cluster buds and the best medium was MS+0.2 mg/L IBA+0.2 mg/L NAA. The rooting in this medium began after 10 d of culture and attained 85.3% rooting rate after 15 d, the plantlet had 4-9 roots in average. The transplant survival rate of plantlets was recorded as 82.3%. [Conclusion]The regeneration system developed in the current experiment will be helpful in commercial scale production of G. Divaricata (L.) DC.关键词
