摘要
Abstract
Aim:To investigate the influence of 5-fluorouracil(5-FU) on the growth and expression of associated genes of pancreatic carcinoma cell sw 1990. Methods;sw 1990 was treated with different concentrations of 5-FU(0,50,100,200, 400 and 800 mg/L)for24, 48,72 h, then was to detect the inhibition ratio by MTT method,and p53,bax,and pten mRNA by RT-PCR and P53 ,BAX,PTEN, and phosphorylated AKT(p-AKT) proteins by Western blot. ResultS:5-FU could inhibit the proliferation of swl990 in a concentration dependent manner ( Fconcentration = 11. 024, P =0.012). 5-FU could increase the expression of p53,bax and pten mRNA in a concentration dependent manner ( Foml:ma,im =4. 317 ,6. 812,and 10.403, P<Q.05;Ftime =3.058,0.906,and0.451,P>0.05).The P53 and BAX protein level in swl990 cells treated with 5-FU was higher and p-AKT protein level was lower than those untreated(t = 9. 025 ,24. 562, and 7.239.P < 0. 05 ) , while the PTEN protein level did not change(t =4. 938,P =0. 845). Conclusion:5-FU may inhibit proliferation of swl990 cells by regulating the expression of p53,bax mRNA and P53,BAX and p-AKT proteins.关键词
