Abstract

The effect of neutron irradiation on somatic embryogenesis in embryonic leaflet culture of peanut(Arachis hypogaea L.)was studied.The embryonic leaflets separated from seeds of peanut cultivar Luhua 11,which were irradiated at dosages of 0,9.7,14.0 and 18.0Gy,were incubated on MSB5 medium containing 10.0mg/L 2,4-D to induce somatic embryos.After 4 weeks culture,the leaflets were transferred to MSB5 medium supplemented with 4.0mg/L BAP.The embryonic leaflets from the unirradiated seeds formed somatic embryos directly on induction medium containing 10.0mg/L 2,4-D.However,after irradiation most of embryonic leaflets first formed calli,when transferred to MSB5 medium containing 4.0mg/L BAP,some of them formed somatic embryos.Only a few of them formed somatic embryos directly on induction medium containing 10.0mg/L 2,4-D.The somatic embryo formation rates varied with different dosages of neutron irradiation.The percentage of somatic embryo formation declined with the increase of neutron irradiation dosages.When the irradiation dosages were increased from 9.7Gy to 14.0 and 18.0Gy,the percentage of somatic embryo formation declined from about 55% to 40% and 30%,respectively.The unirradiated embryonic leaflets gave a higher rate of somatic embryo formation(above 80%).The results showed that neutron irradiation was efficient to induce genetic mutation in peanut and the optimum dosage was about 14.0Gy.
