Abstract

In order to detect the relativity of single nucleotide polymorphism of MyoG gene and adipo-cyte fatty acid binding protein (A-FABP) gene with meat quality in the geese, the crude fat and some muscle fiber traits were detected, using the techniques of PCR-SSCP and sequencing to study the single nucleotide polymorphism of exon 1 of MyoG gene and intron 2 of A-FABP of Taihu geese, and then relativity a-nalysis was conducted. The results showed; (1) The PCR products of exon 1 of MyoG gene had about 420 bp, the mutational site was on the 64 bp (C→T). There was a significant difference of muscle fiber density and muscle fiber diameter between different genotypes(P<0. 05). (2) The products of PCR had about 482 bp, the intron 2 of A-FABP was from 174 bp to 394 bp, there were three mutational sites, respectively on the 187 bp (C→T), 235 bp (C→A) and 372 bp(A→C); (3) By using PCR-SSCP ,two genotypes of the intron 2 of A-FABP were found, there had some difference between different genotypes. Compared the sequence of intron 2 of goose with duck and chicken, the homology were 91% and 85%, respectively. These findings laid good theoretical foundation for the progressive research of the relationship between genetic mutation in MyoG and A-FABP gene with meat trait and for the progressive research of the application of molecular markers in animal breeding.
