Abstract

Objective The present study investigates the difference in the protein expression profiles of drug-resistant HCT-8/VCR cells of human colon carcinomas with and without curcumin treatment Methods The drug-resistant HCT-8/VCR cells of human colon carcinomas were cultured with curcumin at a dosage of 25 μmol/L for 24 h. and those in the control group were treated with physiological saline. Then, the total proteins of both groups were extracted. Solid-phase pH-gradient-based 2-D gel electrophoresis (2-DE) was used to separate the total protein in the drug-resistant HCT-8/VCR cells of human colon carcinomas with and without curcumin treatment, and the gels were stained with silver. A GS-800 scanner was used to produce the gel images, and the differentially expressed proteins were identified and analyzed using the PDQuest 8.0 software program. Results The 2-DE HCT-8/VCR cell patterns were acquired with clear background, high resolution, and better reproduction. Furthermore, the average protein spots of the images with and without curcumin treatment were 1030+69 and 1070 + 96. respectively, hi the curcumin-treated group, 33 protein spots were found to be differentially expressed, 8 were upregulated, and 18 were downregulated. Moreover, only two protein spots were expressed in the curcumin-treated HCT-8/VCR cells, whereas five were found in the control group. Conclusions The 2-DE profiles of human colon carcinoma HCT-8/VCR cells with and without curcumin treatment were primarily established, and 33 differentially expressed protein spots were distinguished. Further studies on these differentially expressed protein spots may be useful in understanding the mechanism of curcumin in reversing multidrug-resistant colon carcinoma cells.
