抗菌肽Folicidin-3在大肠杆菌中融合表达及抑菌活性
Biological characteristics of the antibacterial peptide Fowlicidin-3 fusion expressed in Escherichia coli

Abstract
Based on the amino acids of Fowlicidin-3 and the preference condon of Escherichia coli, a 117 bp Fowlicidin-3a gene and a 102 bp Fowlicidin-3b gene were designed and synthesized. The two genes were cloned to the-express vector to construct recombinant plasmids pET-32a-C-Fowlicidin-3a and pET-32a-Tev-C-Fowlicidin-3b. Then recombinant plas-mids were transferred into E. Coli BL21 Rosetta-2 and induced by IPTG. The fusion proteins Trx-Fowlicidin-3a and Trx-Fowlicidin-3b were expressed and purified. The results showed that Fowlicidin-Sa and Fowlicidin-3b were effectively obtained by cleavage of the fusion protein with enterokinase and Tev proteases. Antimicrobial activity assays demonstrated that the Fowlicidin-3a and Fowlicidin-3b had candidacidal activities. The minimal inhibitory concentration of Fowlicidin-3a and Fowlicidin-3b was 2 μg · mL-1.关键词
