摘要

Abstract

This research selected ten different types of Brassica oleracea L. Var. Italica as test materials, and studied the genotype types,childhood microspores period of cytology observation,culture medium type and pH value etc in isolated microspore culture system of Brassica oleracea L. Var. Italica. The test results showed that, donor plant genotype was the main factor for Brassica oleracea L. Var. Italica microspore embryo thallose yield, and the embryonic induction frequency was difference in different varieties. The optimal length of buds in different genotypes also had certain differences. There was a relationship between the buds and the cytological stage of microspores in Brassica oleracea L. Var. Italica and the suitable of buds had difference in different genotypes. 1/2 NLN+130 g sugar was the best culture medium of the isolated microspore; the best of the culture medium pH value was 6. 2. The suitable incubation time of the cotyledons embryos in liquid medium was 25 days. In MS medium under the appropriate agarose concentration(7.0g·L-1 ) condition the survival rates of embryo thallose seedlings was highest.
