Abstract

High-affinity copper transporter (COPT) is the key member of CTR/COPT family, which plays an important role in mediating copper homeostasis in organisms. Using the primers designed from the known EST from a suppression subtractive hybridization -Cdna library, the full -length Cdna sequence of HbCOPT5 was cloned from Hevea brasiliensis latex by 3-RACE technique. The opening reading frame(ORF)of HbCOPT5 was 420 bp in length, encoding 139 amino acids. HbCOPT5 was predicted to contain three transmembrane domains (TMDs) with molecular weight 15.67 ku and theoretical Pi 5.26. Phylogenetic reconstruction revealed that HbCOPT5 was more closely related to the COPTs from Ricinus communis and Populus trichocarpa, and most closely related to the Arabidopsis COPT5 among the AtCOPTl-AtCOPT6 transporter family. In addition, HbCOPTS exhibited 2.8 times higher transcriptional expression level in healthy trees than in tapping panel dryness trees.
