Abstract

Dehydration survival rate of PLB pretreated with H20 and ABA solution respectively, as well as physiological indices of PLB before and after desiccation were, preliminarily studied to reveal the effects of ABA solution pretreatment on the dehydration tolerance and it's physiological bases of Phalaenopsis PLB (protocorm-like body). The result showed that: PLB pretreated with 80 jimol/L ABA had obviously lower dehydration rate and very significantly higher survival rate after desiccation. There were not significant difference in dry weight, relative conductivity and survival rate after desiccation between fresh PLB, PLB pretreated with H2O and ABA solution respectively. Pretreatment of PLB with ABA solution could very significantly increase the contents of soluble protein, heat-stable protein, soluble carbohydrate and sucrose, respectively. Also, ABA could very significantly increase SOD activity of PLB and very significantly decrease the activitives of POD and CAT, but increase very significantly the total antioxidative capability of PLB. It is suggested that pretreatment of Phalaenopsis PLB with ABA solution in a short time could not change weigh indices and damage on PLB, and ABA could increase the content of protective substances and increase the total antioxidative capability, and that was the bases of elevating dehydration tolerance of PLB.
