胆木组织培养与快速繁殖技术研究

Abstract
Tissue culture and rapid propagation of Nauclea officinalis were conducted with stem segments, leaves, and petioles of in vitro seedlings. Results demonstrated that the best treatment of seed surface sterilization was dipping in 70% alchol solution for 10 s, followed by dipping in 2% solution of the hypochlorite for 8-10 mim. The optima] media for multiple shoots induction and multiplication were 1/2 WPM + 0.5mg/L 2, 4-D, followed by 1/2 WPM+0.5 mg/L IBA and 1/2WPM+1.0 mg/L IBA+0.5 mg/L 6-BA. The multiplication coefficient and the germination rate of shoots was up to 4.8 and 94% respectively. The suitable media for shoot elongation and rooting were(l/2~l/4)WPM +0.5 mg/L 2, 4-D and(l/2-l/4)WPM+0.5 mg/L IBA. The survival rate of plantlets after acclimatization and transfer to glasshouse was approximately 80%.关键词
