As2O3对肾癌细胞增殖、凋亡的影响及意义
Effect and significance of arsenic trioxide on the cell proliferation and apoptosis of renal carcinoma

Abstract
Objective To explore the effect and significance of arsenic trioxide( As2O3) on the cell proliferation, ap optosis and expression of XIAP of human renal carcinoma cell lines 786-0. Methods To collect the 786-0 on the logarith mic growth phase, which were effected by 0. 5, 1.0, 2. 0 μmol/L As2O3 respectively, the inhibiting proliferation of the 786-0 all was analyzed by MTT assay; the apoptosis rate of the 786-0 was measured by flow cytometry; the protein expres sion of XIAP and XIAP mRNA level of the cells were detected by the Western blot and RT-PCR respectively. Results 0.5, 1.0, 2.0 μmol/L As2O3 inhibited the 786-0 proliferation and the inhibition rate was increased as the time prolonging (all P < 0.01). With the increase of the arsenic trioxide concentration, the apoptosis rate was increased (all P < 0.01) , the expression of XIAP and XIAP mRNA was obviously decreased (all P <0.05). Conclusions As2O3 can inhibit 786-0 cell proliferation and induce cell apoptosis. The mechanism may be related to the inhibition on the expression of XIAP of human renal carcinoma 786-0.关键词
