摘要

Abstract

Objective To investigate the effect of tetracycline on rat bone mesenchymal stem cells (BMSCs) injury indued by glucocorticosteroid. Methods The model of rat BMSCs injury induced by glucocorticosteroid in vitro was estab lished, and then isolately cultured in vitro. The BMSCs of rats were divided into six groups; normal control group, hormone control group, 4, 20, 100, 500 u,g/ml tetracycline group. The injury of BMSCs induced by the glucocorticosteroid was de tected by Acridine orange-ethidium bromide double staining. The cytotoxicity of BMSCs was measured by MTT assay, and then drawing cell growth curve. Staining and analysis of bcl-2, bax epressions were detected by immunohistochemical. Western blot was used to detect the expression of caspase-3. Results Acridine orange-ethidium bromide double staining showed that the cell damage was apoptosis. Tetracycline had protection effect on cell proliferation of BMSCs inhibited by glucocorticosteroid by dose-dependent mode. 100 μg/ml tetracycline could induce the apoptosis of BMSCs by increasing the bcl-2, decreasing bax、caspas-3. Conclusion Tetracycline can protect the injury of BMSCs induced by glucocorticoste roid. This provide some experimental pharmaco-foundation for intervention therapy of early osteonecrosis.
