Effect of recombinant BIM on apoptosis and BIM expression transfection in NSCLC with EGFR mutation

Abstract
Objective To analyze the effect and the mechanism of the apoptosis in non-small cell lung cancer cell (NSCLC) transfected by recombinant BIM. Methods A549, H1650, H1975 in logarithmic growth phase were divided into three groups randomly, transfect group was transfected by recombinant BIM, negative control group ( NC group) was transfected without recombinant BIM, black control group ( BC group) was not transfected. After 48 h, apoptosis of cell strains transfected by recombinant BIM was measured by Annexin PI; expression of BIM transfected by recombinant BIM was detected by western blot assay. Results In contrast with NC and BC groups, the apoptosis rate and the expression level of transfect group increased observably, and which were higher in the H1650 and H1975 mutant cell lines than those in the A549 cells (P <0. 01). Conclusion Recombinant BIM transfection can induce the apoptosis in NSCLC cells with EGFR mutations, which possibly caused by increasing the expression of BIM; this provides basis for the gene therapy of NSCLC.关键词
