Biological activities of a synthetic antimicrobial peptide derived from the cathelicidin family

Abstract
Objective To study the effect of synthetic antimianrobial peptide derived from the Cathelicidin family on staphyoccocus aureus. Methods According to the method of NCCLS, the minimal inhibitory concentration ( MIC) and the minimal bactericidal concentration ( MBC) of the synthetic antimicrobial peptide against staphyoccocus aureus were deter mined. Peptide was incubated with a freshly prepared erythrocyte suspension from human blood (5% of normal hematocrit) and released hemoglobin which was determined photometrically at 540 nm. The peptide (80 μg/ml) was added to an over night culture of staphyoccocus aureus. This culture was placed at 37 ℃ and sampled every hour for 4 h and viable bacterial numbers were determined. Results The MIC was 16 μg/ml and the MBC was 128 μg/ml. The peptide exhibited hemolyt-ic activity below 10% at 100 μg/ml. After 4 h the vible bacteria in group A (antibacterial peptide) was significantly de creased compared with that in group C (control) (P<0.05). Conclusion The synthetic antimicrobial peptide possesses obvious anti-bacterial effect on staphyoccocus aureus and displays less hemolytic activity.关键词
