Abstract

Objective To study the effect of synthetic antimianrobial peptide derived from the Cathelicidin family on staphyoccocus aureus. Methods According to the method of NCCLS, the minimal inhibitory concentration ( MIC) and the minimal bactericidal concentration ( MBC) of the synthetic antimicrobial peptide against staphyoccocus aureus were deter mined. Peptide was incubated with a freshly prepared erythrocyte suspension from human blood (5% of normal hematocrit) and released hemoglobin which was determined photometrically at 540 nm. The peptide (80 μg/ml) was added to an over night culture of staphyoccocus aureus. This culture was placed at 37 ℃ and sampled every hour for 4 h and viable bacterial numbers were determined. Results The MIC was 16 μg/ml and the MBC was 128 μg/ml. The peptide exhibited hemolyt-ic activity below 10% at 100 μg/ml. After 4 h the vible bacteria in group A (antibacterial peptide) was significantly de creased compared with that in group C (control) (P<0.05). Conclusion The synthetic antimicrobial peptide possesses obvious anti-bacterial effect on staphyoccocus aureus and displays less hemolytic activity.
