Abstract

Objective To investigate the effects of shenfu injection (SFI) on the induction of chronic myeloid leuke mia cell line K-562 cells into dendritic cells (DCs). Method In vitro, K-562 cells were cultured with different concen trations SFI, granulocyte-macrophage colony-stimulating factor ( GM-CSF)/interleukin-4 (IL-4) and GM-CSF/IL-4 com bined with different concentrations of SFI, respectively. Results In vitro, DCs could not be derived from K-562 cells cul tured with different concentrations of SFI. While DCs derived from K-562 cells co-cultured with GM-CSF/IL-4 could ex press phenotype of antigen-presenting cells and have the similar immune function with normal DCs, but the surface markers and the function were unmature. However, different concentration of SFI plus GM-CSF/IL-4 could not only induce matura tion of K-562 DCs but inhibit propagation of cells. Cells cultured with middle concentration SFI (75 mg/ml) plus GM-CSF/IL-4 expressed higher levels of phenotypes, elicited significantly stronger abilities of stimulating T lymphocyte proliferation, and secreted larger amounts of IL-12. Conclusions In vitro, appropriate concentrations of SFI plus GM-CSF/IL-4 can induce differentiation and maturation of K-562 DCs, change the expression of their surface markers and enchance their immune stimulatory function. The best concentration of SFI is 75 mg/ml which plus GM-CSF/IL-4 can induce K-562 cells into DCs.
