摘要

Abstract

Objective:To study the expression of chemerin in rats kupffer cells(KCs) of hepatic ischemia-reperfusion injury(IRI) and the effects of resolvin El(RvEi) on it. Methods: seventy-two SD rats divided were randomly into three groups: the sham operation group (control group) , the ischemia-reperfusion group (IRI group) and the RvEi treatment group (RvEi group). A model of partial warm ischemia/reperfusion injury in the rat liver was established. RvEi were given ten minutes before the operation. One hour, six hours, twelve hours and twenty-four hours after the reperfusion, Blood was taken from inferior vena cava to test the hepatic enzyme level, including ALT and AST. Kupffer cells (KCs) were isolated and incubated. Tumor necrosis factor alpha (TNF-a)and interleukin-lbeta (IL-1J3) in the supernatants were measured by enzyme linked immunosorbent assay(ELISA). The expression of chemerin in KCs was detected by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR). Results: The ALT and AST level of the RvEi group were significantly lower than those in the ischemia/ reperfusion group(P<0. 01). The mRNA and protein expression of chemerin and the level of TNF-o and IL-l|3in the IRI group significantly increased in the course of the reperfusion and slightly decreased at 24 h, but were still significantly higher than those in the control group (P<0. 05). The expression of these factors were markedly decreased after RvEi treatment compared with the IRI group (P<0. 05). Conclusion: RvEi protect against ischemia/reperfu-sion injury of the rats liver partially by restraining the KCs activation, reducing the excess release of TNF-a and IL-1(3 from KCs and decreasing the high expression of chemerin protein and mRNA.
