摘要

Abstract

Objective To investigate the influence of fragile x mental retardation-1 (FMR1) gene on the spermatogenesis and reproduction of male mice. Methods FMR1 knockout (KO) male mice and wild type (WT) male mice were mated with wt female mice. The number of litters, pregnancy rate and male mice having offsprings were counted. Serum T, FSH and LH concentrations were also measured. The density, mortality and morphology of the left cauda epididymis sperms were analyzed, HE staining was performed on the right side. Results The pregnance rate of wt female mice mated with FMR1K0 males was significantly lower than the control group (41.7% vs. 87. 5% , P < 0.05) ,the average litter size was 6.50 ± 2.27 and 8.22 ± 3.03 (P > 0.05). Male fertility showed that 41.7% of KO mice had pups, whereas 91.7% of the mice had pups in the control group (P < 0.05). No obvious pathological difference in testes and epididymides was found between the two groups. There was no significant difference in the sperm count, motility and abnormal sperm ratio in two groups. Conclusions FMR1 gene may play an important role in the development of male genital system and the absence of FMR1 reduce the reproduction of male mice. The mechanisms of FMR1 gene influencing the male genital system are needed to further study.
