Expression of epithelial-to-mesenchymal transition related proteins in human esophageal cancer and its clinical significance

Abstract
Objective To investigate the expression of epithelial-to-mesenchymal transition ( EMT) related protein markers, E-cadherin, Cytokeratin and Vimentin, in human esophageal cancer tissues and its clinical significance. Methods The immunohistochemistry assay and tissue microarray (TMA) technology were used in the study. 105 cases of esophageal cancer tissues and 11 cases of normal esophageal tissues were involved in the immunohistochemical staining of E-cadherin, Cytokeratin and Vimentin. The correlation of the immunostaining intensities of these makers in esophageal cancer tissues and patients' clinicopathological parameters was analyzed. Results According to the immunohistochemical score system, in 105 cases of esophageal cancer tissues, there were 63 cases with higher E-cadherin expression,42 cases with lower E-cadherin expression;and 51 cases with higher Cytokeratin expression,54 cases with lower Cytokeratin expression;and 35 cases with higher Vimentin expression,70 cases with lower Vimentin expression. The E-cadherin expression level in cancer tissues ( H-score: 172. 7 ± 40. 7) was significantly higher than that in normal tissues ( H-score :31. 9±33.1)(P<0. 0001). The Cytokeratin expression level in the tumor size ≥ 3. 5 cm group was significantly lower than that in the tumor size < 3. 5 cm group (P = 0. 0137). Moreover, in the higher E-cadherin expression group, the Vimentin expression level was significantly lower than that in lower E-cadherin expression group (χ2 =4. 464,P = 0. 0346) . Conclusions The expression variation of EMT related proteins in the oncogenesis and progression of esophageal cancer indicates that the EMT issue involves in esophageal cancer tissues, and the further investigation of EMT in esophageal cancer would help to illustrate the molecular mechanism of invasion and metastasis in this malignancy.关键词
