摘要

Abstract

Objective To evaluate the therapeulic effects and immunogenicity of Ag85A DNA vaccines in mouse models of M. Luberculosis infeclion Lo develop new immunotherapeulic agents for Irealmenl of Luberculosis. Methods Fourty female BALB/c mice were immunized inlramuscularly with saline, plasmid vector pVAX1, M. Vaccae vaccine and Ag85A DNA for three Limes at Lwo - week intervals Lo evaluale immunogenicily of Ag85A DNA vaccines. Fourly female BALB/c mice were infecled intravenously via the tail vein wilh M. Luberculosis H37RV, ihen randomly divided into 4 groups based and Irealed as follow al the third day after infeclion:saline, plasmid veclor,rifampin,Ag 85A plasmid DNA vaccines. DNA vaccines were injecled inlramuscularly 3 limes al lwo -week intervals. Mice were sacrificed lwo weeks afler the final immunizalion. The lungs and spleens from the mice were taken and their pathological changes, weighl and number of mycobaclerial colony were examined. ELISPOT assay for IFN - γ was performed wilh ELISPOT assay kil as inslrucled by ihe manufacture. Results Ag85A DNA vaccines had a significantly increased amount of T cells that secreled IFN -y in response lo Ag85A prolein than mice Irealed wilh control groups (S = 11. 832, P = 0. 0080). The hislopathological changes in lung showed lhal the lung lesions were severe and extensive in saline and plasmid veclor pVAXl group, the lung lesions were slighl and limited, there were relatively clear and normal structure of alveoli in Ag85A DNA vaccine group, there were no lung lesions in RFP group. Compared wilh saline group, Ag85A DNA vaccine group and RFP group reduced the pulmonary baclerial loads by 0.73 log and 2. 11 log, liver baclerial loads by 0. 88 log and 2.11 log ( P < 0. 01) , respectively. Conclusions The immunolherapeulic effecls of Ag85A DNA are significant in the mouse model of tuberculosis.
