Abstract
Objective To investigate the effective relationship between the MicroRNA-21 (miR-21) down-regulation and the biological behaviour and FTEN expression change in gastric cancer, and the possible mechanisms of miR-21 in gastric cancer initiation and development. Methods Transfect the gastric cancer cell lines that miR-21 high expression validated previously by the research team with miR-21 inhibitor; the influence on the proliferation, apoptosis, cell cycle and migration in gastric cancer cell lines were evaluated by CCK-8 kit and Annexin-V+PI and Transwell test; and to evaluate the change of PTEN expression after miR-21 depression in gastric cancer cell lines with Western blotting and luciferase reporter assay. Results The status of cell proliferation in vitro showed that the down-regulation of miR-21 produced an obvious inhibitive effect on the gastric cancer cell proliferation (P<0.05); in vitro experiments also showed that miR-21 depression promoted apoptosis of gastric cancer cell lines of miR-21 high expression (P<0.05), mainly G1/S in the cell cycle (P<0.05); Transwell test showed that cell migration were notablely inhibited (P<0.05); the Western blotting and luciferase reporter assay also showed that PTEN expression increased remarkably after miR-21 inhibition (P< 0.05). Conclusions Down-regulation of miR-21 has a strong inhibitory effect on proliferation in the gastric cancer cell lines of miR-21 high expression, and promotes apoptosis of these cells; meanwhile, depresses migration in vitro; and miR-21 down-regulation up-regulates the PTEN expression level; so PTEN may be a target gene in gastric cancer initiation and development.关键词
