Abstract
Objective:To investigate the therapeutic effect of the acetoacetate extract of Vitex Negundo Seed( Evn-50) and four fractions of Lignans from Evn-50(Comp 6,Comp 7, Comp 8,and Comp 10) on human ovarian cancer COC1 cell line. Methods: COC1 cells that cultured in vitro were divided into five experimental groups ( EG) and three control groups (CG). Ells ere treated with different concentrations of Evn-50,Comp 6,Comp 7,Comp 8, Comp 10 and control drug. The try pan blue exclusion method was used to investigate the growth inhibitory effect of Evn-50 and four fractopms of Lignans of Vitex negundo seed on COC1 cells. The established tumor xenograft in nude mouse model was used to evaluate therapeutic effect of Evn-50 and four fractions of Lignans on human ovarian cancer. Results: Evn-50 and Comp 6, Comp 7,Comp 8,Comp 10 all inhibited viability of COC1 cells significantly in a dose-dependent manner and a time -dependent manner in vitro. Comp 8 was the strongest one with IC50 value of 1. 40μg/ml. Evn-50 was the second one with IC50 of 1.56 μg/ml. The tumor growth inhibitory effect of Evn-50 and Comp 6 at a dose of 5mg/kg were 57. 1 % and 55. 7% respectively. The tumor weight inhibitory rates were 61. 6% and 53. 1% respectively. Comp 8 had little effect. Conclusion: The acetoacetate extract of Vitex negundo seed ( Evn-50 ) and the Lignans from the acetoacetate extract of Vitex negundo seed have an inhibiting effect on the growth of human ovarian cancer COC1 cell line both in vitro and in vivo.关键词
