Optimization of Tissue Culture in Xinjiang White Garlic by Using Quadratic Regression Orthogonal Rotation Combination Design

Abstract
[Objective] The purpose of this project aimed to study the effects of the plant hormones on Xinjiang white garlic bulb disadventitious shoots growth in order to provide a theoretical basis for rapid propagation system. [ Method ] The experiment adopted quadratic regression orthogonal rotation combination design, design of different IB A, NAA,6 - BA level and their combination. [Result] The result showed that the primary and secondary sequence of the factors that affect adventitious shoots growth is: NAA > 6 - BA > IBA, among which, the interaction between NAA and IBA was significant but with negative effects, so in the highest concentration prediction, IBA is removed to reduce interaction to Xinjiang white garlic bulb disc adventitious shoots growth effects. The single 6 - BA has more obvious effects (usually positive effect) than other factors. [ Conclusion ] The best ratio of hormone for Xinjiang white garlic bulb disc adventitious shoots growth was; NAA concentration was 2 mg/L, 6 - BA concentration was 7. 5 mg/L. In this condition Xinjiang white garlic disc adventitious shoots growth can reach the maximum, which was 39.22.关键词
