猪ApoA5基因cDNA的克隆、序列分析及组织表达研究
Molecular Cloning, Sequence Analysis and Tissue Expression of Pig ApoA5 Gene

Abstract
This experiment was conducted to obtain and analyze cDNA sequence of pig Apoli-poprotein A5 gene (ApoA5) , and to study the tissue expression characterization of ApoA5 mR-NA in pigs. By reverse transcription PCR and rapid amplification of cDNA ends (RACE), the full-length cDNA of ApoA5 was cloned from liver of Mashen pig, and the expression of ApoAb mRNA in different tissues of Mashen and Large White pigs were examined by quantitative realtime PCR. The results showed that the cDNA of pig ApoA5 contained 1 917 nucleotides, including a 1 092 bp open reading frame (ORF) flanked by 24 bp 5' UTR and 801 bp 3' UTR. Pig ApoA5 encoded 364 amino acid residues, which shared 81. 7%, 80. 0%, 78. 3%, 77. 5%, 76. 5% , 73. 6% , 67. 1% and 66. 7% identity with those of Bos taurus , Equus caballus , Homo sapiens , Canis familiaris , Macaca mulatta , Oryctolagus cuniculus , Rattus novegicus and Mus musculus , respectively. The expression level of ApoA5 mRNA was higher in liver, medium insubcutaneous adipose and longissimus dor si, and lower in small intestine, kidney, heart, pancreas and stomach. But no expression was detected in spleen and lung. The expression of ApoA5 mRNA in subcutaneous adipose and longissimus dorsi exhibits different patterns between the two pig breeds. The result indicate that the ApoA5 gene is highly conservative during the course of animal evolution. The expression of ApoA5 mRNA is influenced by tissue and breed factors. It was inferred that ApoA5 gene may be related to adipogenesis and adipose metabolism.关键词
