磺胺二甲嘧啶快速直接竞争ELISA试剂盒的研制及应用
Development and Application of Rapid Direct Competitive ELISA Kit for Sulfamethazine
摘要
Abstract
The aim of this study was to develop the direct ELISA kits based on polyclonal antibody to detect the sulfamethazine in pork liver, pork and chicken, and the results were confirmed by HPLC. Sulfamethazine was spiked to pork liver, pork and chicken and was detected by the ELISA and HPLC. The sensitivity, specificity, precision, veracity and validity of the kits were verified. Its detection limit was 0. 47 ng · mL-1 with a concentration range from 0. 5 to 50 ng · mL-1. The mean intra-assay variability and inter-assay was 7. 4% and 9. 27%, respectively. The detection limits were 3. 91, 4. 81 and 1. 59 μg · kg-1 in pork liver, pork and chicken. The recovery was between 85%-110% when spiked to samples at 10. 0-50. 0 μg · kg-1 , which is adaptable to the veterinary use. The kit's detection time is 12. 6 min, and can be stored at 4 °C at least 6 months. The kit is satisfying rapid screen of SMZ residue in pork liver, pork and chicken.关键词
