Abstract
Objective To discuss the feasibility of using mitochondrial CO1 gene as DNA bar code to identify species of murid. Methods The genome DNA was extracted from 42 murid samples collected in the Sino - Nepali border area. The mitochondrial C01 gene was amplified by method of consensus primer PCR and sequenced. BLAST comparison was made between the sequencing results and the DNA bar codes of other murid species in genebank,and molecular evolution tree was built. Results In this research, specific C01 gene straps in 17 samples were amplified by PCR. The average intraspecies genetic distance of the 17 samples was 0.035. The average interspecies genetic distance was 0. 47. The interspecies genetic distance was almost 10 times more than the intraspecies genetic distance. According to the BLAST comparison, the 17 samples were classified in different murid species. Conclusion The C01 gene order can be used effectively to identify the murid species.关键词
