实时定量PCR技术在角膜移植供体乙型肝炎病毒筛查中的应用

Abstract
Objective; To evaluate the feasibility of using real-time quantitative PCR technique in HBV DNA screening for comeal graft material. Methods; Blood sample, extraocular fascia, cornea, pigmented membrane from 22 corneal transplantation donors were qualitatively and quantitatively analyzed with real-time quantitative PCR. The specificity and sensitivity of detecting HBV DNA with blood specimen and ocular tissues were compared, so were the differences of HBV DNA content among ocular tissues. Results; In donative materials from 18 cases with HBV positive and 4 with HBV negative, sensitivities of HBV DNA detection were 83% (15/18), 100% (18/18), 83% (15/18)and 78% (14/18) with blood sample, extraocular fascia, cornea and pigmented membrane, respectively; while specificities were all 100%. HBV DNA quantitative analysis in extraocular fascia, cornea and pigmented membrane showed that unit quality pigmented membrane HBV DNA load was the highest, and no significant difference between those in extraocular fascia and cornea was found. Conclusion; Detection of HBV DNA in extraocular fascia with real-time quantitative PCR technique can be applied in corneal transplant donor pathogen screening.关键词
