食药用真菌粗提物促血管内皮活性的筛选及对高糖损伤内皮的保护作用
Effects of edible-medicinal fungus crude extracts on vascular endothelial cells with high glucose-induced injury
摘要
Abstract
AIM; To invesligale lhe effecls of crude exlracls of Cordyceps gunnii (CGE) , LepisLa lenLinus ( LLE) , Cordyceps sinensis ( CSE) and LenLinus slriguellus ( LSE) on lhe proliferation of high glucose - Lrealed human umbilical vein endolhelial cells ( HUVECs). METHODS: The cullured HUVECs were divided inlo normal conlrol group ( Lrealed wilh M199 cullure medium alone) , high glucose group ( lrealed wilh M199 cullure medium containing 33 mmol/L glucose) and 4 crude exlracls of edible - medicinal fungi ( CGE, LLE, CSE and LSE) intervention groups (lrealed wilh lhe crude exlracl of edible -medicinal fungus al concenlralions of 12. 5, 25, 50, 100 mg/L in high glucose M199 cullure medium). The cell proliferation was evaluated by MTT assay. The cell cycle and ROS level were measured by flow cylome-Iry. RESULTS; Compared wilh normal conlrol group, lhe MTT absorbance value and lhe percentage of G0/G1 slage of lhe HUVECs in high glucose group were significantly decreased ( P < 0. 05 ) , while lhe percentage of S + G2/M and ROS level were significantly increased (P<0. 05). Compared with high glucose group, Ireatmenl with lhe crude exlracls of LepisLa lenLinus and LenLinus sLriguellus decreased lhe cell absorbance value ( P < 0. 05 ) , and the inhibitory effect was enhanced in a dose - dependenl manner. However, Cordyceps gunnii had no effect ( P > 0. 05 ) . The crude extracts of Cordyceps sinensis ( 12. 5 ~50 mg/L) significantly enhanced lhe proliferative activity of HUVECs, decreased the percentage of G0/G1 stage of HUVECs, increased lhe percentage of S + G2/M of HUVECs, and reduced lhe intercellular ROS level (P < 0. 05) . CONCLUSION: Only Cordyceps sinensis crude exlracl effectively prolecls lhe HUVECs in high glucose - induced injury, which mighl be due Lo promoting more cells Lo enler Lo lhe cell cycle and down - regulating oxidative slress.关键词
