Whole genome DNA microarray construction and comparative genomic analysis of Brucella
摘要
Abstract
To develop whole genome DNA microarray based on the genomic of Brucella,a total of 3 218 genes of Br.abortus 16M and Brucella9-941 were downloaded from GenBank for primers design.After several rounds of condition optimization and redesign of primers,PCR products of 3 212 genes were finally obtained,purified and printed onto glass slides to construct a microarray.The microarray includes a total of 6 912 probes,including double spots for each gene,and positive and negative control.Using Cy3/Cy5 double staining the genes of Brucella 16M as reference sample and genes of Br.abortus 9-941 test samples,the comparative genomic hybridization was developed.The microarray hybridization results were perfectly consistent with the theory result.In conclusion,the microarray can be used as a useful tool for comparative genomic analysis of Brucella.关键词
