Abstract

OBJECTIVE: To analyze the inner correlation of in vitro drug susceptibility and multidrug resistance genes MDR1, MRP and LRP with clinical efficacy in acute leukemia patients, and to provide the basis for clinical individualized medication. METHODS: 35 acute leukemia patients were collected from hematology department of our hospital during Feb-Jul. 2010, and ATP-TCA was adopted to detect the in vitro drug susceptibility and RT-PCR was used to detect the multidrug resistance genes MDR1, MRP and LRP. RESULTS: The total coincidence rate of in vitro drug susceptibility and clinical efficacy was 85.7% , the negative accordance rate was 50.0% , the positive accordance rate was 90.3%, the response rate was 93.3% and the specificity was 40.0%. There were 23 cases of MDR1, MRP and LRP negative expression (65.7%) and 2 cases of positive expression (5.7%). The effective rate of MDR1 negative cases was 93.3% and that of positive cases was 40.0% (P<0.01); the effective rate of MRP negative cases was 89.7% and positive cases was 66.7% (P>0.05); the effective rate of LRP negative cases was 92.6% and positive cases was 62.5% (P<0.05). Among 23 cases of in vitro drug susceptibility and negative drug resistance genes MDR1, MRP and LRP, 21 cases were relieved with effective rate of 91.3% (21/23). Among 2 cases of in vitro drug susceptibility and positive drug resistance genes MDR1, MRP and LRP, none of them were relieved with the effective rate of 0 (0/2). CONCLUSION: For acute leukemic patients, simultaneously detection of in vitro drug susceptibility and multidrug resistance genes MDR1, MRP and LRP has certain predicted value, and could be used as laboratory basis for acute leukemic patients personalized medication.
