Abstract

OBJECTIVE: To optimize the formulation of Compound ornidazole chitosan vaginal bioadhesive film. METHODS: The formulation was optimized by orthogonal design with the chitosan concentration, gelatin concentration, PVA concentration and drug concentration as factors using vertical force as index. The bioadhesiveforce and in vitro drug release rate of the films were measured as well. RESULTS: The optimum formulation was as follows: chitosan 2.0%, gelatin 1.0%, PVA 1.0% and drug 2.0%. The average bioadhesiveness of 3 batches of optimized preparations was 2.1 N·cm-2, and the in vitro drug release rate of the films in 12 h was more than 89%. CONCLUSION: The optimal Compound ornidazole chitosan vaginal bioadhesive film shows proper bio-adhesion force and slow-release character.
