摘要

Abstract

OBJECTIVE: To study the protective effects of supercritical CO2 extract from Atractylodes japonica (GCT)against ethanol-induced gastric mucosal injury in rats. METHODS: 40 Wistar rats were divided into 5 groups, I.e. Normal control group (constant volume distilled water) , model group (constant volume distilled water) , hydrotalcite group (0.37 g ? Kg"1) , GCT high-dose and low-dose groups (0.70,0.35 g-kg"1).The rat model of acute gastric damage was induced by absolute ethanol (0.5 mL? 100 g"1) intragastrically. Those groups were given relevant medicine intragastrically once a day for 5 days. Model was induced 30 min after the last medication. The gastric mucosal injury area was observed. The contents of MDA and NO, the activity of SOD, MPO and NOS, and the expression of COX-2 in gastric mucous were determined. RESULTS: High-dose and low-dose of GCT suppressed the elevation of MDA and MPO, and enhanced the SOD activity, the decrease of NO content and NOS activity. Moreover, GCT significantly reduced COX-2 expression. CONCLUSION: GCT can prevent ethanol-induced gastric mucosal injury, partly through scavenging free radical, maintaining NO level and down-regulating COX-2.
