苯溴马隆对罗格列酮在小鼠体内药动学的影响
Effects of Benzbromarone on Pharmacokinetics of Rosiglitazone in Mice

Abstract
OBJECTIVE: To explore the pharmacokinetic characteristics of rosiglitazone in mice, and to investigate the effect of benzbromarone on pharmacokinetics of rosiglitazone. METHODS: Mice were randomized into single drug group (rosiglitazone 0.2 rng·kg-1) and drug combination group (rosiglitazone 0.2 mg·kg-1+benzbromarone 5 mg·kg-1) with 55 mice in each group. Those mice were given relevant medicine intragastrically. The concentration of rosiglitazone in plasma was measured by HPLC-FLU at 0.17, 0.33, 0.75, 1.5, 2.0, 2.5, 4.0, 6.0, 8.0, 12.0 and 24.0 h after medication, and the pharmacokinetic parameters were calculated with 3p97 software. RESULTS: Main pharmacokinetic parameters of rosiglitazone in single drug group vs. Drug combination group were as follows: cmax: 253.1 ng·mL-1 vs. 649.5 ng·mL-1; t1/2Ke:0.94 h vs. 0.74 h; AUC0~24b:511.3 ng·h·mL-1 vs. 1 150.9 ng·h·mL-1. Compared with single drug group, cmax and AUC0~24h of drug combination group increased significantly (P<0.05). CONCLUSION: Benzbromarone can remarkably increase the absorption of rosiglitazone in mice.关键词
